ANALIZA’s LogD, Aqueous Solubility and BBB Permeability analysis use chemiluminescent nitrogen detection as a universal detector. The process is suitable for analysis of compounds provided in DMSO solutions, so long as the compound contains nitrogen. The software uses the compound’s molecular weight (formula weight if a salt) and molar nitrogen content to convert the raw signal into units of mg/mL and LogD, so this information is required for each sample.

Please provide the following:

  • Sample Volume for Samples submitted in DMSO:
    • BBB Permeability: 250µL at 10-30mM concentration in 100% DMSO
    • LogD only: 40µL at 10-30mM concentration in 100% DMSO
    • Aqueous Solubility only: 10µL at 10-30mM solution in 100% DMSO
    • LogD and Aqueous Solubility: 50µL at 10-30mM solution in 100% DMSO
  • Maximum of 90 compounds + 2 wells of DMSO used for dissolution per 96 well plate to accommodate our on-board QC Standards
  • Polypropylene 96 well plates
    • We recommend the following plates and seals for source plates due to their low nitrogen content:
      • Abgene, 1.2mL round bottom polypropylene plate, Part Number: AB-0564
      • Abgene, 200µL conical bottom polypropylene plate, Part Number: AB-1058
      • Abgene Easy Peel Foil Heat Seal: Part Number: AB-0745
      • Corning 300 ul round bottom, Part Number 3365
  • Frozen samples shipped on dry ice
  • Analiza Sample Submission form (Excel Spreadsheet). The completed form should be returned to akestranek@analiza.com prior to shipping samples
  • For Dry Powders, Thermodynamic Solubility
    • Powders should be provided in 2 ml glass vials with Teflon- or silicone-lined septa or cap to avoid leaching of nitrogenous plasticizers into the sample
    • Sample amount should correspond to the desired upper solubility limit, with a ratio of 1 mg of sample for each 1 mg/mL upper solubility limit
    • Analiza will add 1mL of desired solvent to the vial
    • Analiza Sample Submission form (Excel Spreadsheet). The completed form should be returned to akestranek@analiza.com prior to shipping samples.
